Overview

RNA binding motif protein 45 (RBM45) is a highly conserved RNA-binding protein expressed predominantly in the fetal brain and decreasing with development. It contains three RNA recognition motif (RRM) domains, each capable of binding RNA sequences, most notably GAC and GACG motifs, and a central HOA (homo-oligomer assembly) domain responsible for self-association and interaction with other RBPs such as FUS and TDP-43. RBM45 localizes chiefly in the nucleus but can shuttle to the cytoplasm, where it forms aggregates especially in neurons and glia affected by ALS and FTLD. It functions in pre-mRNA splicing by binding to intronic splicing enhancers and is essential for proper maturation of select mRNAs. Abnormalities in RBM45, including mutations and protein aggregation, are implicated in neurodegenerative diseases, but direct therapeutic targeting has not been established.
