Overview

RNA binding motif protein 46 (RBM46) is a germ cell–specific RNA-binding protein characterized by one or more RNA recognition motifs (RRMs)[2][4]. RBM46 is highly expressed in testis and is essential for gametogenesis, particularly in regulating the transition from mitosis to meiosis in germ cells by binding to specific sequences (GCCUAU/GUUCGA motifs) in the 3' untranslated regions (3'UTRs) of mRNAs encoding meiotic cohesin subunits such as SMC1B, SMC3, STAG3, and RAD21[1][3][5]. Loss of RBM46 function results in meiotic arrest and infertility, with its primary role being the post-transcriptional regulation of mRNAs critical for proper chromosome segregation and synaptonemal complex assembly during meiosis[3][5]. While RBM46 is classified as a cancer/testis antigen (CT68), there are no established therapeutics targeting this protein and its principal biological importance remains in reproductive cell differentiation and meiosis, not in established therapeutic targeting[2][5].
