Overview

RNA binding motif protein 5 (RBM5) mRNA encodes a multi-functional RNA-binding protein that acts as a significant tumor suppressor (UniProt P52948). It is located within the 3p21.3 tumor suppressor locus, a region frequently deleted in various malignancies, particularly lung cancers (PubMed 15534194). The primary biological function of RBM5 is the regulation of alternative splicing, where it influences the production of pro-apoptotic isoforms of genes such as FAS and CASP2 (PubMed 22431518). In clinical contexts, decreased levels of RBM5 mRNA are associated with poor prognosis and increased tumor aggressiveness in non-small cell lung cancer and other solid tumors (PubMed 17332323). While there are currently no FDA-approved drugs that directly target RBM5 mRNA, it is a subject of research for RNA-based therapies, including antisense oligonucleotides and gene replacement strategies aimed at restoring its tumor-suppressive functions (PubMed 25650595). Experimental studies have demonstrated that restoring RBM5 expression can effectively inhibit cell proliferation and induce apoptosis in various cancer cell lines (PubMed 11062476).
