Overview

RNA-binding motif protein X-linked-like 2 (RBMXL2) is a nuclear RNA-binding protein in the heterogeneous nuclear ribonucleoprotein (hnRNP) family with two RNA recognition motif (RRM) domains. It is highly conserved in placental mammals and is primarily expressed in male germ cells during meiosis, when it suppresses aberrant splicing by repressing cryptic splice site selection, especially in long or ultra-long exons. RBMXL2 prevents the production of faulty mRNA isoforms that would disrupt protein coding and is essential for meiotic progression and male fertility. Knockout studies in mice show that loss of RBMXL2 leads to defects in germ cell development and complete male infertility. No pharmacological agents or clinical biomarkers are currently associated with this target. Note: RBMXL2 is not currently considered a direct therapeutic target (e.g., for drugs), but it is important for understanding germ cell biology and the molecular basis of certain male infertility syndromes.
