Overview

RNA binding motif single stranded interacting protein 2 (RBMS2) is a member of a small family of proteins that bind single-stranded DNA and RNA[3][5]. The protein contains two ribonucleoprotein consensus sequence domains (RNP-CS with RNP1 and RNP2 motifs), which are necessary for nucleic acid binding[3]. RBMS2 is involved in various core processes, including regulation of gene transcription, alternative splicing, DNA replication, progression of the cell cycle, and apoptosis[1][3]. It is implicated as a tumor suppressor in several cancers—its downregulation is observed in renal clear cell carcinoma, lung cancer, gastric cancer, and hepatocellular carcinoma, where it may contribute to cancer progression[1]. In kidney renal clear cell carcinoma, low RBMS2 expression correlates with more advanced disease and poorer outcomes, suggesting a role as a prognostic biomarker and a potential therapeutic intervention target[1]. Mutations or altered regulation of RBMS2 have also been associated with amyotrophic lateral sclerosis and other diseases[3]. Currently, no specific drugs are reported to directly target RBMS2, nor are there reported mechanisms of action for RBMS2-targeted pharmacology or notable safety/toxicity concerns in therapeutic contexts[3][1].
