Overview

RNA binding motif single stranded interacting protein 3 (RBMS3) is an RNA-binding protein belonging to the c-Myc single-strand binding protein family, characterized by two ribonucleoprotein consensus sequences required for DNA and RNA binding[1][5]. RBMS3 is primarily cytoplasmic and has a key role in stabilizing specific mRNAs, regulating their translation, and modulating various cellular processes—including cell proliferation, cell cycle progression, apoptosis, and post-transcriptional gene regulation[1][2][4][5]. It acts as a tumor suppressor in multiple malignancies by inhibiting oncogenic signaling pathways such as Wnt/β-catenin, suppressing epithelial–mesenchymal transition (EMT), and stabilizing key transcripts like Twist1 and PRRX1[1][2][4]. A reduced expression of RBMS3 is frequently associated with enhanced tumor proliferation, metastasis, chemoresistance, and poor clinical outcomes[1][2][3]. Beyond cancer, RBMS3 is involved in embryonic development (pancreas, neural crest, chondrogenesis), fibrosis, and immune regulation[1][3]. It is a promising prognostic biomarker and a potential therapeutic target in oncology, particularly for aggressive and metastatic cancers, though currently there are no approved drugs directly targeting RBMS3[1][2][4].
