Overview

RNA binding protein fox-1 homolog 2 (RBFOX2) is an evolutionarily conserved RNA-binding protein that regulates the alternative splicing of tissue-specific exons in metazoans, especially within the nervous system but also in other tissues. The crucial feature of RBFOX2 is a highly conserved RNA recognition motif (RRM) domain, which binds specifically to RNA sequences containing the (U)GCAUG element, imparting strict sequence specificity. Through its nuclear localization signal, RBFOX2 predominantly localizes to the nucleus where it modulates splice site selection, impacting the expression of protein isoforms critical for cell differentiation and function. The protein regulates numerous genes, and dysregulation or mutation of RBFOX2 is associated with cancer, developmental, and neurological disorders. No known small molecule drugs directly target RBFOX2. As a splicing factor, it serves more as a molecular regulator than a classic therapeutic target such as a receptor or enzyme.
