RNA binding protein fox-1 homolog 3 (RBFOX3) is a neuron-specific nuclear protein and an essential regulator of alternative pre-mRNA splicing, particularly in postmitotic neurons[1][2][3][4][5]. It contains an RNA recognition motif (RRM) and binds specifically to the UGCAUG element within pre-mRNA, modulating neuron-specific splicing events critical for neural differentiation and function[1][5]. The protein is most widely known as the NeuN antigen, a standard marker for mature neurons in research and pathology. RBFOX3 is highly expressed in the central nervous system and is central to neural development and adult brain function. Mutations or loss of RBFOX3 have been linked to various neurodevelopmental and neuropsychiatric disorders, including epilepsies such as benign epilepsy with centrotemporal spikes, and may play a role in other brain diseases, although it is not considered a therapeutic target. NeuN (RBFOX3) immunoreactivity is used extensively as a biomarker for identifying post-mitotic neurons, but expression can vary with developmental stage, injury, or disease[1][2][3][4][5].
