Overview

RNA-binding protein PNO1 is a highly conserved RNA-binding protein involved in the biogenesis of the small ribosomal subunit in eukaryotes. As a component of the small subunit (SSU) processome, PNO1 facilitates critical steps in ribosome assembly, including rRNA folding, modification, and cleavage, and positively regulates dimethylation of adenosines in the 18S rRNA. Beyond its basic cellular function, PNO1 is upregulated in multiple cancers where it promotes tumor cell proliferation, cell cycle progression, migration, invasion, and survival by engaging in several oncogenic signaling pathways (such as MAPK, TGF-β, and Notch), inhibiting ferroptosis, and regulating autophagy and metabolic reprogramming. Its suppression can reactivate tumor-suppressive pathways like p53. PNO1 is essential for global protein synthesis and is localized primarily in the nucleolus and nucleoplasm[1][3][4].
