Overview

RNA-binding protein with serine-rich domain 1 (RNPS1) is a nucleocytoplasmic shuttling protein encoded by the RNPS1 gene in humans. RNPS1 is a core component of the exon junction complex (EJC), which is recruited to newly spliced mRNA. It participates in mRNA nuclear export, mRNA surveillance, and especially in nonsense-mediated mRNA decay (NMD), a quality control mechanism targeting mRNAs with premature stop codons for degradation. RNPS1 is also involved in both constitutive and substrate-dependent alternative splicing, often modulating the expression of apoptotic genes such as Bcl-X. It forms part of larger multiprotein complexes (e.g., ASAP and PSAP) that transiently associate with the EJC during pre- and post-splicing mRNA processing. RNPS1 is not typically considered a classical therapeutic target such as a receptor or enzyme, but its dysregulation is associated with cancer and certain neurodegenerative and skeletal diseases[1][2][8].
