Overview

The RNA component of 7SK nuclear ribonucleoprotein (RN7SK, commonly "7SK") is a highly abundant, non-coding small nuclear RNA present in metazoan cells. It forms a ribonucleoprotein complex (7SK snRNP) with core proteins including HEXIM1/2, LARP7, MePCE, and P-TEFb (CDK9/CyclinT1). 7SK RNA regulates transcription by sequestering and inactivating P-TEFb, a major RNA polymerase II kinase, thus inhibiting transcription elongation. The complex undergoes conformational changes depending on protein interactions and cellular signals. Dysregulation of RN7SK or associated proteins is linked to various diseases such as cancer, cardiovascular conditions, and viral infections. Despite its essential role in transcription regulation, RN7SK is not directly targeted by approved drugs. Its regulatory network is a subject of intense study due to its central role in controlling gene expression and cell fate.
