Overview

RNA exonuclease 1 homolog (REXO1) is a eukaryotic enzyme with predicted 3'-to-5' exonuclease activity, implicated mainly in rRNA 3'-end processing and found in the nuclear body[1][3][4]. REXO1 encodes a large protein (1221 amino acids), containing an exonuclease domain, and can bind Elongin, a transcription elongation factor[5]. While REXO1 does not appear to directly affect transcription elongation in vitro, it is recognized as a marker for embryonic stem cells and is expressed in both embryonic and multipotent stem cells as well as certain cancer lines[1][5]. Its presence is tightly regulated at the epigenetic level and lost upon cell differentiation, demarcating pluripotent from lineage-primed states[1]. There are no known small-molecule drugs or therapies that directly target REXO1, nor are there established safety concerns specific to its targeting.
