RNA incorporation of pyrimidine analogs refers to the process by which synthetic or modified pyrimidine nucleoside analogs are mistakenly incorporated into RNA molecules during transcription, in place of their natural counterparts (cytidine and uridine). This misincorporation can disrupt normal RNA function and is a key mechanism underlying the cytotoxic effects of several chemotherapeutic agents, most notably 5-fluorouracil (5-FU). This also represents an emerging strategy for antibiotic development due to its high barrier for resistance evolution among pathogens.
