LEO1 is a conserved protein subunit of the RNA polymerase II-associated factor 1 complex (PAF1C), which is involved in the regulation of eukaryotic gene transcription. As a member of PAF1C, LEO1 participates in transcription elongation, mediates histone modification and nucleosome turnover, and plays roles in maintaining open chromatin architecture. LEO1, often as a heterodimer with Paf1, contributes to both the modulation of chromatin state and the DNA damage response by helping target regulatory complexes and facilitating removal of stalled RNA polymerase II from sites of DNA damage. While critical for gene expression control, LEO1 is not recognized as a receptor, enzyme, or direct drug target.
