Overview

RNA polymerase II elongation factor ELL (ELL) is an essential transcription factor that enhances the processivity of RNA polymerase II during transcriptional elongation by suppressing transient pausing and facilitating promoter clearance and pause site entry/release[1][2][3][4]. ELL is a central component of the so-called super elongation complex (SEC), which promotes high-amplitude gene induction and participates early in the transcription cycle to stabilize polymerase recruitment and chromatin structure[2]. ELL is also clinically significant due to its involvement in the pathogenesis of leukemia through fusion events with the MLL (KMT2A) gene, forming oncoproteins that deregulate transcription and are associated with aggressive disease[3]. ELL’s activity is vital to development and gene regulation, with knockout or mutation studies in model organisms showing lethality and disrupted gene expression[3]. While the protein itself is not the direct target of approved drugs, its fusion forms and transcriptional complex roles are of considerable interest in cancer therapeutic research.
