Overview

RNA polymerase II-mediated transcription machinery is a highly conserved molecular system in eukaryotes, essential for synthesis of messenger RNAs and certain noncoding RNAs. The machinery consists of the 12-subunit RNA polymerase II enzyme and auxiliary transcription factors—such as TFIID, TFIIH, and Mediator—that assemble in large macromolecular complexes to regulate genome-wide transcription. Proper coordination of initiation, pausing, and elongation steps by this machinery underlies the precise control of gene expression. Disruptions or alterations in machinery components are implicated in diverse diseases, including cancer, developmental disorders, and infections. Therapeutic targeting is possible but limited by toxicity and complexity of the machinery
