Overview

RNA polymerase III subunit GL (POLR3GL) is a protein-coding gene in humans that encodes a specific peripheral subunit of the RNA polymerase III complex—a multi-protein enzyme responsible for transcribing small non-coding RNAs including tRNAs, 5S rRNA, snRNAs, Y RNAs, and vault RNAs. POLR3GL is the gene for the RPC7β subunit, which is mutually exclusive with its paralog, POLR3G (encoding RPC7α), and is broadly expressed in differentiated tissues, in contrast to POLR3G, which is enriched in embryonic stem cells and tumor cells. While POLR3GL and POLR3G are functionally redundant in most contexts, their differential expression provides a mechanism for controlling RNA polymerase III transcriptional activity during development, differentiation, and disease. Disruption of POLR3GL is associated with developmental abnormalities including short stature with multiple congenital defects, and is also essential for viability in mammals. The protein helps assemble the Pol III complex and is involved in transcription initiation, but not elongation or termination. There are currently no known drugs directly targeting POLR3GL, and its main disease relevance is as a potential biomarker and essential enzyme component.
