Overview

RNA pseudouridine synthase domain containing 1 (RPUSD1) is a protein-coding enzyme predicted to catalyze pseudouridine synthase activity—specifically, the isomerization of uridine to pseudouridine in ribosomal RNA (rRNA)[1]. RPUSD1 is part of the larger family of pseudouridine synthases, which modify RNA by converting uridine bases to pseudouridine, a process crucial for stabilizing RNA conformations and influencing ribosome structure and function[2]. RPUSD1, like other pseudouridine synthases, contains an RNA-binding domain and a highly conserved catalytic aspartate necessary for its enzymatic activity[2]. These enzymes play essential roles in ribosome biogenesis, RNA maturation, and translation fidelity. No direct small molecule inhibitors, clinical drugs, or routine biomarkers are currently reported for RPUSD1, and its roles in human disease remain unclear; however, similar enzymes have been linked to defects in protein synthesis and cellular disease phenotypes[2].
