Overview

The RNA synthesis machinery encompasses the molecular components responsible for transcribing DNA into RNA. The core enzyme is RNA polymerase, with eukaryotic cells possessing three main types: RNA Polymerase I (rRNA synthesis), RNA Polymerase II (mRNA and some snRNA synthesis), and RNA Polymerase III (tRNA, 5S rRNA, and other small RNA synthesis). These polymerases are multi-subunit complexes requiring accessory factors for proper function. The machinery catalyzes RNA strand formation complementary to DNA, involving initiation, elongation, and termination. Products are essential for gene expression, with mRNAs serving as templates for protein synthesis, rRNAs forming ribosomes, and tRNAs delivering amino acids. Errors can lead to diseases like Diamond Blackfan anemia or Dyskeratosis congenita. Many antibiotics target bacterial ribosomal RNAs, inhibiting protein production, highlighting the machinery's importance as a drug target.
