Overview

Ro60 RNA-binding protein is an evolutionarily conserved ring-shaped autoantigen found in most animal cells, parts of prokaryotic genomes, and some archaea. It forms ribonucleoprotein complexes with noncoding Y RNAs, binding specifically to a conserved stem structure in Y RNA. Ro60 is central in RNA quality control, stabilizing and binding Y RNAs and misfolded or aberrant RNAs. In prokaryotes, Ro60 homologs cooperate with exonucleases to enhance RNA degradation. Ro60's role is crucial in maintaining RNA integrity, and deficiencies sensitize cells to environmental stresses, such as UV irradiation. In humans, Ro60 is the target of anti-Ro/SSA autoantibodies, implicated in several autoimmune diseases, and its associated RNP complex is pivotal for both cellular RNA metabolism and the initiation/pathogenesis of systemic autoimmune rheumatic diseases. This summary is based on authoritative reviews and structural studies. There is no evidence Ro60 is directly modulated by any drugs, but its clinical significance as an autoantigen is well-established.
