Rod-derived cone viability factor long isoform (RdCVFL) is an intracellular thioredoxin enzyme encoded by *NXNL1*. It plays a crucial role in maintaining retinal health through antioxidant defense mechanisms that protect cone photoreceptors from degeneration—especially relevant in diseases like retinitis pigmentosa where secondary loss of cones leads to blindness. Unlike its short secreted counterpart (RdCVF, which acts extracellularly), RdCVFL’s action is cell-autonomous, making it a promising target for neuroprotective therapies aimed at preserving vision after rod cell death.
RdCVFL reduces disulfide bonds in target proteins, modulating their activity and protecting cells from oxidative stress. It interacts with metabolic enzymes like PKM2 to restore glycolytic flux under oxidative stress conditions, promoting cell survival.
