Overview

RORA antisense RNA 2 (RORA-AS2) is a long non-coding antisense RNA transcribed from the RORA locus, on the strand opposite the coding gene for retinoic acid receptor-related orphan receptor A (RORA). Antisense RNAs like RORA-AS2 can regulate the expression of their target sense gene (in this case, RORA), typically by transcriptional interference, RNA-RNA pairing leading to mRNA degradation, or influencing chromatin organization in cis or trans[2]. Although antisense RNAs are important in regulatory gene networks and are emerging as tools/targets for therapy (mainly through synthetic antisense oligonucleotides), there is currently no evidence that RORA-AS2 itself serves as a direct therapeutic target, nor that it has any direct implication as a disease biomarker or target molecule in human disease. The primary functional relevance of RORA-AS2 would be indirect regulation of RORA, a nuclear receptor and transcription factor with pleiotropic roles in metabolism, cardiovascular and immune functions, inflammation, and cancer[1][3][5].
