Overview

Rotavirus G1 reassortant is a genetically engineered viral strain used as a primary immunogen in live-attenuated vaccines, most notably the pentavalent RotaTeq (RV5). It is constructed using a bovine rotavirus (WC3) backbone that expresses the human VP7 surface glycoprotein specific to the G1 serotype, which is a dominant cause of severe pediatric gastroenteritis worldwide (CDC, 2021; FDA, 2020). The biological function of this reassortant is to mimic natural infection at the intestinal mucosa to stimulate the production of neutralizing IgA antibodies without causing clinical disease (NCBI, 2020). By targeting the VP7 protein, the vaccine induces serotype-specific immunity that prevents viral attachment and entry into enterocytes. While highly effective in reducing hospitalizations due to rotavirus, the use of this reassortant is associated with a small but significant risk of intussusception, necessitating careful post-marketing surveillance (StatPearls, 2023). It serves as a critical tool in global public health efforts to mitigate the burden of diarrheal mortality in children.
