Rotavirus A VP7 is a major outer capsid glycoprotein that plays a critical role in the viral life cycle and pathogenesis. It is the primary determinant of the G-serotype (glycoprotein-type) and is essential for the attachment and entry of the virus into host intestinal epithelial cells (enterocytes). Specifically, VP7 forms the smooth outer surface of the triple-layered rotavirus particle and is involved in the calcium-dependent stability of the capsid. In the context of disease, Rotavirus A is the leading cause of severe dehydrating diarrhea in infants and young children worldwide. Serotype G3 is one of the most prevalent genotypes globally, making it a key target for vaccine development. Therapeutic intervention primarily involves live-attenuated vaccines, such as RotaTeq and Rotasiil, which include the G3 serotype to elicit protective neutralizing antibodies that block viral infection. Research also explores monoclonal antibodies and subunit vaccines targeting VP7 for passive immunization and broader protection.
