Overview

Rotavirus G3 reassortant is a live, attenuated virus strain used as a key component in pentavalent rotavirus vaccines like RotaTeq (FDA, 2021). It is engineered by reassorting the gene encoding the VP7 glycoprotein from a human G3 rotavirus strain into a bovine rotavirus (WC3) genetic backbone (Matthijnssens et al., 2008). The VP7 protein is a primary target for neutralizing antibodies, and the G3 serotype is a major contributor to severe diarrheal disease in children globally (CDC, 2023). Upon oral administration, the reassortant replicates in the small intestine and induces a mucosal and systemic immune response (WHO, 2021). This provides protection against subsequent infections by wild-type G3 rotaviruses, significantly reducing the incidence of severe gastroenteritis and related hospitalizations (Greenberg & Estes, 2009).

Mechanism of action

The G3 reassortant induces active immunity by presenting the human G3 VP7 antigen to the host immune system, leading to the production of serotype-specific neutralizing antibodies and memory B cells (FDA, 2021; WHO, 2021).
