Rotavirus surface antigen VP4 is a spike-forming protein located on the outer capsid of rotaviruses. It mediates the virus's attachment to host epithelial cell receptors and facilitates viral entry through membrane penetration. VP4 is cleaved into VP8* (receptor binding) and VP5* (membrane penetration) subunits. It is a key target for vaccine development due to its ability to induce strong mucosal and systemic immune responses. VP4 also contains an actin-binding domain that promotes actin cytoskeleton remodeling for nonlytic virus exit.
