Rotavirus surface proteins, specifically VP4 and VP7, are the primary antigenic components of the live-attenuated Rotarix vaccine strain (RIX4414). VP7 is a glycoprotein that forms the outer shell of the virus, while VP4 forms spikes that facilitate attachment to host cells. In the context of the Rotarix vaccine, these proteins serve as the targets for the host's immune system to generate neutralizing antibodies. These antibodies prevent subsequent infection by wild-type rotavirus by blocking viral entry and uncoating in the intestinal epithelium. Rotavirus is a leading cause of severe diarrhea and dehydration in infants and young children worldwide. By presenting these surface proteins in an attenuated form, the vaccine prepares the immune system to recognize and neutralize various rotavirus serotypes, primarily those sharing G1 and P[8] specificity.
Induction of neutralizing antibodies (primarily IgA and IgG) that target the VP4 and VP7 surface proteins of the rotavirus, thereby blocking viral attachment to host cells and preventing subsequent infection.
