VP4 is an outer capsid spike protein of Rotavirus that mediates attachment to host cell receptors and facilitates membrane penetration during entry. It undergoes tryptic cleavage into VP8* and VP5*. VP8* binds sialic acid on host cells and VP5* is involved in membrane permeabilization. It also contains an actin-binding domain (RV-ABD) that interacts with actin cytoskeleton to promote virus exit.
