Overview

Rotavirus VP7 G2 glycoprotein is a major structural component of the outer capsid of Group A Rotaviruses, specifically those belonging to the G2 genotype (UniProt P04502). It is a 37-kDa calcium-binding protein that forms the smooth outer shell of the triple-layered infectious particle (PMID: 18462440). VP7 plays a critical role in viral entry by facilitating the attachment and subsequent penetration of the host cell membrane (PMID: 25824474). As one of the two primary antigens—along with VP4—that define the viral serotype and induce neutralizing antibodies, it is a central target for vaccine development (CDC, 2023). The G2 genotype is frequently associated with the P[4] genotype and is a common cause of severe diarrhea in infants worldwide. Vaccines like RotaTeq specifically include a G2 reassortant to provide immunity against this strain, while others rely on cross-neutralization (PMID: 17484215). Monitoring variations in the VP7 G2 protein is essential for tracking vaccine efficacy and the emergence of escape mutants. The protein's stability is highly dependent on calcium concentrations, and its loss triggers the uncoating process necessary for viral replication (PMID: 12610144).
