Overview

Rotavirus VP7 glycoprotein (G1 serotype) is a major structural protein that constitutes the outer shell of the rotavirus virion (UniProt: P04502). It functions as a critical mediator of viral attachment and entry into host enterocytes within the small intestine, facilitating the delivery of the viral core into the cytoplasm (PubMed: 15105533). The G1 serotype is one of the most epidemiologically significant genotypes, frequently associated with severe diarrhea and dehydration in infants and young children worldwide (WHO). Because it is a primary target for neutralizing antibodies, the G1 antigen is a core component of major commercial vaccines, such as Rotarix and RotaTeq (FDA). These vaccines work by eliciting a protective immune response, primarily through the production of intestinal IgA, which prevents subsequent infection by G1-type viruses (PubMed: 22438551). Monitoring the genetic stability and antigenic drift of the G1 VP7 protein is vital for ensuring the long-term efficacy of global immunization programs.

Mechanism of action

Vaccines containing or expressing the G1 antigen induce the production of neutralizing antibodies (primarily IgA) that target the VP7 protein, preventing viral attachment and entry into host enterocytes (PubMed: 15105533, CDC).
