Roundabout guidance receptor 2 (ROBO2) is a transmembrane protein that functions primarily as a receptor for the SLIT family of proteins, especially SLIT2. It plays a crucial role in axon guidance and cell migration during nervous system development. In vertebrates, it prevents axons from recrossing the midline by responding to Slit ligands secreted by floor plate cells at the CNS midline. Beyond initial wiring, ROBO2 also contributes to lateral pathway formation, cell migration, synaptogenesis, and maintenance of inhibitory synapses in adult brain regions. Alterations or mutations affecting ROBO2 have been implicated in several neurodevelopmental disorders.
