Overview

Roundabout guidance receptor 1 (ROBO1) is a single-pass transmembrane cell surface receptor that belongs to the immunoglobulin superfamily and functions primarily as an axon guidance receptor by mediating cellular repulsive responses to the secreted SLIT-family ligands. ROBO1 plays a key role in directing axons within the central nervous system by regulating their crossing at the midline and also contributes to cell migration, adhesion, angiogenesis, and organ development. In humans, ROBO1 has been linked to neurodevelopmental disorders such as dyslexia and is implicated in cancer biology, notably in tumor angiogenesis and progression. While direct therapeutic modulation is not clinically established, ROBO1 and its signaling pathway are under investigation as both targets and biomarkers in oncology and neurodevelopmental research.
