Roundabout homolog 1 receptor (ROBO1)

Target
ROBO1
Molecular classification
Receptor, Cell adhesion molecule, Axon guidance receptor, Member of the immunoglobulin superfamily
01

Overview

Roundabout homolog 1 receptor (ROBO1) is a transmembrane protein encoded by the *ROBO1* gene. It belongs to the immunoglobulin superfamily and functions primarily as an axon guidance receptor that mediates cellular responses to molecular cues—most notably through binding SLIT family ligands—to regulate neuronal navigation across the midline during central nervous system development. In addition to its critical role in neural patterning and cell adhesion, ROBO1 has been implicated in regulating cell proliferation and migration outside of neural tissues. Clinically, genetic variants or disruptions of *ROBO1* have been associated with neurodevelopmental disorders such as dyslexia and may influence susceptibility to certain cancers through pathways affecting cellular growth control[1][2][3][5].

Mechanism of action

(There are no approved drugs targeting this molecule; however, experimental modulation would likely involve altering axon guidance or cell migration by interfering with Slit–ROBO signaling.)

03

Biological functions

Axon guidance and midline crossing in the central nervous systemCell migration, especially neuronal migration during developmentSignal transduction for repulsive axonal cues (notably via Slit ligands)Regulation of cell proliferation in some contexts, such as cancer
04

Disease associations

Neurodevelopmental disorders (e.g., dyslexia, other communication disorders)Cancer (e.g., pancreatic cancer proliferation inhibition)Hirschsprung disease (via altered expression with its ligand SLIT2)
