Overview

Roundabout homolog 4 receptor is a transmembrane protein encoded by the ROBO4 gene in humans. It acts as a receptor for Slit proteins—at least for SLIT2—and plays a critical role in angiogenesis and vascular patterning, processes essential for the development and maintenance of the circulatory system. ROBO4 is primarily expressed in endothelial cells, where it contributes to the establishment of the endothelial barrier. It has been shown to suppress glioma-induced endothelial cell proliferation, migration, and tube formation by inhibiting VEGFR2-mediated activation of PI3K/AKT and FAK signaling pathways. The molecule is also known as "Magic roundabout" due to its unique role in vascular biology. Its involvement in pathological angiogenesis links it to diseases such as cancer, particularly those involving abnormal blood vessel growth or function.
