Overview

RUN and FYVE domain-containing protein 1 (RUFY1) is an adaptor protein that contains a RUN domain (important for interaction with small GTPases of the Rab and Rap families) and a FYVE-type zinc finger domain (which binds specifically to phosphatidylinositol-3-phosphate [PI3P]). RUFY1 regulates early endosomal trafficking, facilitator of endosome tethering and fusion, and retrieval of proteins from endosomes to the trans-Golgi network through interactions with the dynein-dynactin complex. RUFY1 has functional links to neurodegenerative diseases, cancer, and immune regulation, primarily due to its role in orchestrating cytoskeletal and vesicular dynamics[1][3][4][5]. Its dysfunction or altered expression can impact cellular homeostasis and contribute to pathological processes, particularly those involving impaired trafficking, autophagy, and cell migration. RUFY1 is one member of a small family of RUN and FYVE domain-containing proteins involved in similar trafficking and signaling functions.
