RUN and FYVE domain-containing protein 3 (RUFY3) is a member of the RUFY protein family, defined by an N-terminal RUN domain and a C-terminal FYVE domain in some isoforms, with the classic neuronal isoform lacking the FYVE domain[1][2]. RUFY3 plays crucial roles in endosomes and lysosome positioning, organelle trafficking, cytoskeletal rearrangements, axonogenesis, and cellular migration, particularly in neuronal and immune cells. Alternative splicing yields distinct tissue-specific isoforms: the neuronal (nRUFY3) version involved in axonal growth and polarity, and the immune-specific (iRUFY3) version that regulates endo-lysosomal positioning and immune cell migration[1]. Dysregulated RUFY3 expression is implicated in cancer cell motility and neurodevelopmental processes, but its direct involvement as a therapeutic target is primarily mechanistic, modulating processes rather than acting as a receptor or enzyme directly targeted by drugs[1][2][3].
