Overview

RUN and FYVE domain-containing protein 4 (RUFY4) is a member of the RUFY protein family characterized by a RUN domain, several coiled-coil motifs, and a FYVE domain, and is involved in the regulation of autophagy, endosomal trafficking, and organelle positioning in immune cells[1][3]. RUFY4 acts as an effector for the small GTPase RAB7, facilitating autophagosome maturation and autophagic flux, and is also implicated in the selective clearance of mitochondria (mitophagy) and the elimination of intracellular pathogens by activated macrophages and dendritic cells[1][3]. RUFY4 expression is high in alveolar macrophages and dendritic cells, and its function is regulated by immune stimuli such as Toll-like receptor agonists and cytokines[3][5]. In osteoclasts, RUFY4 enhances bone resorption but suppresses osteoclast differentiation, acting through modulation of cytoskeletal structures and vesicular trafficking processes critical for bone remodeling[5]. No direct drugs or biomarker roles are currently attributed to RUFY4, and while its modulation affects immune and bone homeostasis, no notable safety concerns are documented in the literature. RUFY4 is not conventionally recognized as a classic therapeutic target such as a receptor or enzyme, but rather as an adaptor protein critical for cellular trafficking and innate immune responses[1][2][3][5].
