Overview

RUNX1 antisense RNA 1 (RUNX1-AS1) is a long non-coding RNA (lncRNA) transcribed from the opposite strand of the RUNX1 gene locus. As an antisense RNA, it does not encode a protein but regulates gene expression at the transcriptional and epigenetic levels. RUNX1-AS1 can modulate the expression of RUNX1, a critical transcription factor in hematopoiesis and cancer, and has been implicated in promoting proliferation, migration, and survival in various cancers. Its expression is upregulated in many tumors, including hematological malignancies, and experimental studies suggest RUNX1-AS1 plays oncogenic roles by acting as a molecular sponge for microRNAs, modifying chromatin, or regulating transcription factor recruitment[7]. RUNX1-AS1 is under active investigation as a therapeutic target and biomarker for malignancies, but there are currently no clinically approved drugs that directly target this lncRNA. Key distinctions: - RUNX1-AS1 is a non-coding RNA and not a receptor, enzyme, or traditional druggable protein, but it is considered a viable *non-protein coding therapeutic target* in cancer biology research. - It is distinct from RUNX1 (a protein-coding gene and transcription factor); RUNX1-AS1 modulates RUNX1 mRNA levels and downstream effects[7].

Mechanism of action

Experimental gene silencing or modulation acts through: - Antisense oligonucleotides or siRNA, reducing RUNX1-AS1 expression in cancer cells to inhibit proliferation and induce apoptosis[7]. - Potential synthetic lethal or co-targeting strategies in oncological applications (in preclinical or research settings).

Safety considerations

Specific safety challenges are not yet well-defined in clinical use, as RUNX1-AS1-targeted therapies are preclinical[7].Potential safety concerns may include off-target effects and delivery-related toxicity common to RNA-based therapeutics.
06

Interacting drugs

No FDA-approved drugs currently target RUNX1-AS1 directly
07

Biomarkers

RUNX1-AS1 levels themselves have been proposed as biomarkers for diagnosis, prognosis, or monitoring in certain cancers, such as acute myeloid leukemia and other malignancies[7].Expression status may stratify patients for targeted therapies or disease risk.
