The ryanodine receptor (RyR) is a giant, homotetrameric intracellular calcium release channel primarily located on the membranes of the sarcoplasmic reticulum (SR) in muscle cells and the endoplasmic reticulum (ER) in other cell types. It mediates rapid release of calcium ions from internal stores into the cytosol, a critical step for excitation-contraction coupling in muscles. RyRs are regulated by numerous factors including ions, small molecules, and proteins, and their dysregulation underlies several disorders like malignant hyperthermia and catecholaminergic polymorphic ventricular tachycardia.
