Overview

S-phase kinase-associated protein 2 (SKP2) is an F-box protein and a key component of the SCF E3 ubiquitin ligase complex. It plays a central role in cell cycle regulation, particularly in promoting the transition from G1 to S phase by targeting specific cell cycle inhibitors, such as p27^Kip1^, for degradation. SKP2 acts as an oncogene in multiple solid tumors and hematological malignancies, with overexpression associated with enhanced tumorigenic potential and poor prognosis. It is also involved in metabolic regulation and apoptosis. SKP2 is being explored as a therapeutic target, and small-molecule inhibitors are under investigation for anti-cancer therapy development.
