Overview

S-phase upregulated non-coding 1 (SUNO1) is a polyadenylated, long intergenic non-coding RNA (lincRNA) located on human chromosome 19, expressed at elevated levels during S-phase and G1/S transition of the cell cycle, especially in cancer cells[1][2]. It predominantly localizes to the nucleus as discrete puncta, and depletion (by siRNA or CRISPR knockout) results in defective S-phase entry, reduced cell proliferation, cell-cycle arrest at G1/S, and hypersensitivity to DNA damage. SUNO1 acts by facilitating the cell-cycle-specific transcription of WTIP, which is a positive regulator of YAP1 in the Hippo signaling pathway. Loss of SUNO1 disrupts loading of the MCM complex and impairs DNA replication origin licensing, emphasizing its vital role in cell division and genome integrity[1][2]. There is currently no evidence for pharmacological targeting, clinical use as a biomarker, or known aliases beyond its gene/probe identifiers.
