Overview

S100 calcium-binding protein A2 (S100A2) is a member of the S100 family of low molecular weight, EF-hand calcium-binding proteins primarily expressed in human lungs, prostate, kidney, hair follicles, and secretory glands[1]. S100A2 is unusual among S100 proteins for its predominant nuclear localization but can also be found in the cytoplasm, with its subcellular distribution influenced by calcium concentrations[1]. S100A2 is important for cytoskeleton organization and is implicated in cell differentiation, tissue regeneration, and wound healing; it is induced by the tumor suppressor p53 and participates in p21 transcription[1]. Biochemically, S100A2 functions as a homodimer and, in multimeric forms, can act as a ligand for the receptor for advanced glycation end products (RAGE)[1]. Clinically, S100A2 is notable for lowered expression in various cancers—lung, kidney, prostate, melanoma, and breast—which is associated with poor prognosis and increased likelihood of metastasis[1]. S100A2 does not have established drug interactions or direct therapeutic targeting, but its altered expression makes it a candidate biomarker for certain cancers[1].
