Overview

S100A4 mRNA is the messenger RNA transcript encoding the S100A4 protein, a member of the S100 calcium-binding protein family (NCBI Gene ID: 6275) [NCBI Gene]. S100A4, also known as Metastasin-1 (Mts1) or Fibroblast-specific protein 1 (FSP1), is a key regulator of cell motility and is strongly associated with the metastatic potential of cancer cells (UniProt P26447) [UniProt]. The mRNA transcript serves as a therapeutic target for RNA-based modalities, such as small interfering RNAs (siRNAs) and antisense oligonucleotides (ASOs), which aim to prevent the translation of the S100A4 protein [Boye K, et al., Am J Pathol, 2010]. By silencing S100A4 at the mRNA level, these therapies can inhibit epithelial-mesenchymal transition (EMT) and reduce the invasive capacity of tumor cells [Zhang J, et al., Biomolecules, 2020]. Additionally, S100A4 mRNA is a target in fibrotic diseases, where its downregulation can prevent the activation of fibroblasts into myofibroblasts [Klingelhöfer J, et al., Methods Mol Biol, 2009]. Current research focuses on optimizing the delivery of these RNA-targeting agents to specific tissues to minimize off-target effects and enhance therapeutic efficacy [Fei F, et al., Med Oncol, 2017].
