Overview

S100 calcium-binding protein Z (S100Z) is a member of the S100 protein family, a group of small, EF-hand, calcium-binding proteins expressed in vertebrates[1][2]. Most S100 proteins are cellular signals modulators, responding to changes in intracellular calcium concentrations and interacting with a diverse range of protein partners, including enzymes, cytoskeletal proteins, transcription factors, and receptors[1][2]. While S100Z shares the general architecture and binding properties of the family, its specific expression, partner proteins, and functional roles remain unclear because it is one of the least studied S100 proteins. So far, there is no evidence of therapeutic targeting, direct disease association, or utility as a biomarker for S100Z. S100 proteins broadly regulate processes such as cell cycle, apoptosis, differentiation, and motility, and several family members play extracellular roles via signaling pathways such as RAGE and TLRs, but direct evidence for S100Z’s involvement in these functions is currently lacking[1][2].
