Overview

Sabinene synthase is a plant-specific enzyme belonging to the terpene synthase (TPS) family, primarily responsible for the biosynthesis of the monoterpene sabinene from geranyl diphosphate [UniProt, PubMed: 8144450]. It is found in a variety of aromatic and medicinal plants, such as Salvia officinalis (sage), Citrus limon, and Piper nigrum (black pepper), where it plays a critical role in producing essential oils [PubMed: 12535069]. Sabinene is highly valued in the fragrance and flavor industries for its woody and spicy organoleptic properties and is also involved in plant defense mechanisms against pathogens and pests [PubMed: 22763391]. From a pharmacological perspective, Sabinene synthase is not a recognized therapeutic target in human medicine, as humans lack the metabolic pathways for monoterpene synthesis and no corresponding human homologs exist. There are currently no clinical drugs, therapeutic candidates, or biomarkers associated with this enzyme in the context of human disease treatment [DrugBank]. Current research is largely focused on agricultural biotechnology and metabolic engineering for the sustainable production of natural compounds.

Mechanism of action

Sabinene synthase catalyzes the conversion of geranyl diphosphate (GPP) into the bicyclic monoterpene sabinene (EC 4.2.3.110). This enzymatic reaction involves the ionization of the substrate to a terpinen-4-yl carbocation intermediate, followed by cyclization and deprotonation [KEGG, UniProt]. As this is a plant-specific enzyme with no human homolog, it does not serve as a mechanism of action for therapeutic drugs.
