SAC3 domain-containing protein 1 (SAC3D1) is a protein that plays a crucial role in centrosome replication and spindle formation, both essential processes for accurate cell cycle progression and mitotic division. It is localized to the centrosome, nucleus, and spindle, and functions in the negative regulation of receptor signaling via the JAK-STAT pathway and in regulating the immune response. SAC3D1 is strongly implicated in cancer biology—particularly in hepatocellular carcinoma—where its expression is positively correlated with cell cycle gene expression and cell proliferation. Downregulation of SAC3D1 impairs cancer cell proliferation, migration, invasion, and induces cell cycle arrest and late apoptosis. Nitidine chloride (NC) directly binds to SAC3D1 and suppresses its expression, suggesting SAC3D1 is a promising therapeutic target in tumorigenesis, especially for drug development strategies aiming to regulate the cell cycle in cancer cells.
Downregulation induces cell cycle arrest (S and G2/M phase), induces late apoptosis in tumor cells, and inhibits proliferation, migration, and invasion of cancer cells.
