The sacral nerves are a set of five pairs of spinal nerves (S1-S5) that emerge from the sacral region of the vertebral column and form part of the sacral plexus (StatPearls, 2023). They provide critical motor and sensory innervation to the lower extremities, the perineum, and the pelvic viscera, including the bladder and rectum (NIH, 2024). While the nerves themselves are anatomical structures rather than individual molecular targets, they are the site of action for sacral neuromodulation (SNM), a specialized therapy used to treat refractory overactive bladder and fecal incontinence (Mayo Clinic, 2023). In pharmacological contexts, sacral nerves are common sites for regional anesthesia where local anesthetics like lidocaine are applied to block voltage-gated sodium channels, thereby preventing pain transmission (Wikipedia, 2024). Because this entry represents a complex anatomical bundle of neurons and connective tissue rather than a specific protein or receptor, it is generally classified as an anatomical target rather than a molecular therapeutic target (Cleveland Clinic, 2023).
Local anesthetics inhibit voltage-gated sodium channels on the axons of the sacral nerves to block signal conduction; therapeutic neuromodulation uses electrical stimulation to modify nerve activity.
