Overview

SACS antisense RNA 1 (SACS-AS1) is a long non-coding RNA (lncRNA) transcribed from the SACS locus, oriented antisense to the SACS gene, and does not encode a protein. Antisense RNAs such as SACS-AS1 participate in regulating gene expression at multiple levels, including transcription, splicing, chromatin remodeling, and post-transcriptional regulation[1][2]. They may act in cis—affecting their neighboring sense gene by overlapping or proximity—or in trans, targeting distant loci or mRNAs or acting as competing endogenous RNAs (ceRNAs) to bind microRNAs, thereby influencing gene expression indirectly[1][2]. Many lncRNAs and antisense RNAs, depending on their genomic position and interactions, modulate chromatin state and gene responsiveness, or alter splicing by masking splice sites[1][2][3]. SACS-AS1 specifically has not been characterized as a therapeutic target, as current evidence does not link it directly to disease, drugs, or established biomarker roles.
