Sad1 and UNC84 domain-containing protein 3 (SUN3) is a nuclear envelope protein, part of the SUN domain protein family, serving as a component of the LINC (LInker of Nucleoskeleton and Cytoskeleton) complex in humans[5][3]. SUN3 mediates physical connections between the nuclear lamina and the cytoskeleton, thereby playing a crucial role in transmitting mechanical forces across the nuclear envelope, positioning the nucleus, and nuclear remodeling—especially prominent during the formation of sperm heads in spermatogenesis[5][1][3]. Its proper localization and expression are specifically interdependent with that of another SUN domain protein, SUN4, and SUN3 is believed to stabilize nuclear envelope associations in the male germline[1][5]. Defects in SUN3 or its related complexes have been linked with spermatogenic failure[5]. No evidence exists for direct drug targeting, human disease biomarker utility, or notable safety concerns to date.
