Overview

Salmonella enterica serovar Typhi (S. Typhi) outer membrane and flagellar protein antigens are essential surface components of the bacterium responsible for typhoid fever. These antigens, primarily the flagellar H antigen (flagellin) and various outer membrane proteins (OMPs) such as OmpC and OmpF, are critical for bacterial motility, environmental sensing, and host cell invasion (StatPearls, 2023). In medical science, they serve as the primary targets for the development of typhoid vaccines and diagnostic serological tests, such as the Widal test (WHO, 2023). Vaccines targeting these antigens, including live-attenuated and conjugate forms, work by stimulating the host's immune system to produce neutralizing antibodies and memory cells (PubMed, 2018). These antigens are highly immunogenic, making them ideal for eliciting protective responses against systemic infection. Understanding the structural variability of these proteins is also crucial for monitoring antibiotic resistance and strain evolution in endemic regions (UniProt, 2024).
